November 4, 1576: In Antwerp, Spanish soldiers, having failed to receive their pay, decide to get compensation by sacking the city. The event, known as the Spanish Fury, costs their lives to 7000 civilians and unites the provinces of the Netherlands against Spanish King Philip II's rule, reigniting the revolt that will eventually result in the Netherlands's independence. It also precipitates a major decline in the cloth trade Antwerp was famous for. The city will never completely recover its commercial dominance.

November 6, 1928: Arnold Rothstein, famous for his rigging of the White Sox game in 1919, was shot and mortally wounded on November 4, and died on November 6. He was dragged out of a casino and shot in an alleyway by George "Hump" McManus after failing to pay a gambling debt.

It is kind of funny that this is coming up, because I had listened to something on NPR just recently where they were talking about just that hypothetical situations.
They actually said it was very likely to have not happened if not for the assassination. I don't remember the specifics any more, but they said a lot of things were actually getting better and the assassination started a lot of problems that weren't there before.
Granted there obviously would have been some conflicts between some of the key players of the war anyway, but probably not in the way that it played out.

Indeed this is not the intent of the thread to do a debate on this, but if you're interested in the matter, I'll point you to this excellent book. It doesn't propose a definitive answer, but it convincingly explains how better leadership could have avoided a conflict even after the assassination, while describing how the events since ~1900 had made a war much more likely in the first place.
