I definitely must watch again. I missed some dialog near the middle because the person behind me was blowing her nose constantly. Not a healthy sound at all.

Her laugh was just awesome. Infectious even, I think near the end I was mentally laughing along with her. The crazed smiles she'd have when she laughed made it all the more awesome.

The repeated dosing was quite savvy on her part.
Underestimating Spike could be attributed to what Twilight said to him:
Spike: "Humdrum is just the useless sidekick."
Twi: "Well then it's a good thing you're not Humdrum."
In other words, Mane-iac possibly didn't know Spike's competency. It doesn't seem like she knew they were from outside the comic book (but that's hard to tell since she's completely nuckin bucks).
