No, it isn't. V's cape stays on all the time. You can see the small curved lines from it in the bottom of the panels featuring V.

The fact that V is still wearing the cape here might be a clue too.

In the second, the curve is there, but the red isn't.

The red is there in the book, but V is positioned about 1/10th mm lower in the third to last panel.

If you go backwards (comic 664, 663) you'll see that V's cape is not shown above him, so why should it be in 665? I don't know what your pixel peeping is really revealing, but it seems clear that Roy's makeshift shift is indeed the cape. What other red cloth with the same writing as V's cape do they have?

And by 667 it had simply been returned to V, Roy had clearly given it up by then.

I suppose it could be a 2nd cape if V carries spare clothing.

I randomly decided to say the sound effects out loud and realized that the phylactery makes a different sound when it hits the shield as it is a different material than the last two items

In that strip, Elan thinks he can see V's hump. This would make sense for, as a wizard V spends 8 hours a day leaning over a book studying...or it her lady lumps

The clasp is shown above V as a curve touching the intersection between the contours of the hair and the arm, and can be seen in all the mentioned comics. Yes, it is pixel peeping, but it shows that the clasp, which is a vital part of the cape, stays on all the time, and thus the rest of the cape should do so too.
